Rich, take a look at my 12/21 post on this board. Quite a bit depends on how much you wish to spend.
As for timing, I've had good fishing there in January, February, March, April and May. I have not been to Los Roques at other times but hear that the bone fishing is good year round. It will be May for my son and me this year.
If you do a Google search on Los Roques you'll find links to lodging - posadas - and fishing. I stand by my recommendation of Pez Raton (they book through Frontiers Travel)as the best of the lot.
